Abstract
The quantification of proteins, using isotope dilution LC-MS/MS and Multiple Reaction Monitoring (MRM) of specific peptides from the protein of interest, requires multiple complex sample preparation steps to obtain accurate and reproducible results.
The optimal sample preparation is protein and peptide specific and includes steps for protein denaturation, reduction/alkylation of disulfide bonds, enzymatic hydrolysis and peptide fractionation prior to the LC MS analysis. The complexity of the sample matrix has great influence on these steps, and is very different for PK/TK serum samples, protein manufacturing samples, cell lysis samples, milk samples, etc. The matrix may for example contain thousands of other proteins in serum samples, detergents and additives in manufacturing samples, or fat, phospholipids and fragments from cell lysis samples.
Alphalyse has as CRO gained experience from a variety of protein LC-MS quantification projects, some of which have been validated according to EMEA guidelines for clinical use. Examples will be presented for quantification of protein vaccines, monoclonal antibodies, and therapeutic proteins, as well as troubleshooting and practical experiences gained from different projects.
