Determination of protein electrophoretic pattern by 1D PAGE and 2D PAGE

Do you need separation of proteins by 1D PAGE or 2d SDS PAGE?

We offer to separate the proteins of your drug sample. This is done by either 1D PAGE or 2D PAGE (polyacrylamide gel electrophoresis). We use Coomassie, silver or fluorescent staining for visualization of all proteins in the sample.

Do you follow the ICH Q6B? (Test Procedures and Acceptance Criteria for Biotechnological/Biological Products). The ICH Q6B guideline requires that you determine the electrophoretic pattern of your protein by 1D and 2D gel electrophoresis.

Our service includes:

  • 1D gel electrophoresis or 2d gel electrophoresis separation of your sample.
  • Coomassie, silver or fluorescence staining of the proteins.
  • For 1-D SDS PAGE gel we will load 1-30 ug of protein on gel.
  • For 2-D SDS PAGE we will load 30-300 ug of protein on gel.

1D Gels:
Protein separation by 1D SDS-PAGE is performed using the NuPAGE pre-cast gel system from Thermo Scientific according to the manufactures manual. The protein sample (5 – 20 ug ) is dissolved in LDS sample buffer with DTT and heated (70 oC, 10 minutes), loaded onto the NuPAGE 12% Bis-Tris gel and separated with a MOPS running buffer at 200V for 40-50 minutes. The gel is stained by Coomassie brilliant Blue R250, or by Silver staining (Alphalyse procedure).

 

2D Gel:
2D-SDS PAGE is performed using IPG drystrips 7 cm, pH 3-10 for the first dimension isoelectric focusing, and NuPAGE pre-cast gel system from Invitrogen for the second dimension of Mw separation. The sample is purified/desalted using 2-D Clean-Up Kit (GE Healthcare). The purified sample is mixed with Destreak Rehydration solution with IPG-buffer prior isoelectric focusing. SeeBlue Plus2 Pre-Stained Standard (Invitrogen) is used as marker.

  1. The isoelectric focusing is performed until approximately 8000 Vhrs
  2. The second dimension is performed in NuPAGE precast 4-12% Bis-Tris zoom gels and proteins separated with a MES running buffer at 200V for 45 minutes.

The gels are stained by Coomassie Blue (Merck), or by Silver staining (Alphalyse procedure).

The samples delivered for both 1D SDS PAGE and 2 D PAGE should be delivered in-solution or as a solid material.

For 1d gel electrophoresis we will typically load between 5 and 20 µg and for 2D gels between 50-200 µg.

Ship your protein samples by a courier company of your own choice

Place your samples into micro tubes and pack in a suitable transport box.

  • Contact a courier company of your choice (FedEx, UPS, DHL, TNT, World Courier etc.). Use the following sentence to describe the content: “Protein material – Non-toxic, non-hazardous, for research purpose only“.
  • Use cool packs or cooling units if your samples should be stored below 5°C/40°F
  • Ship the samples to the office closest to you (Canadian customers, please ship to Europe):

Order a prepaid FedEx Air Waybill < 1 lb./ 500g

A prepaid FedEx Air Waybill can be used for fast and easy shipment of samples in packages up to 1 lb./ 500g.
We will generate a prepaid Air Waybill, prefilled with the correct information, and send it to you by e-mail (PDF-format) within 24 hours.
Then follow these simple instructions:

  • Place your samples into micro tubes. Pack in suitable transport box.
  • Print the PDF document and stick it on the transport box or just hand it to your FedEx officer upon pickup.
  • Call to request a pickup from your local FedEx office.

If you plan to send your samples stored at room temperature, this option is usually the right choice. If you fill your box with cooling packs, the weight will most often exceed 1 lb./500g.

Order a prepaid FedEx Air Waybill < 10 lb./5kg (e.g. for packages with cooling packs)

This prepaid FedEx Air Waybill can be used for shipment of samples in packages up to 10 lb./ 5kg, with or without cooling packs (do not use dry ice!).
We will generate a prepaid Air Waybill, prefilled with the correct information, and send it to you by e-mail (PDF-format) within 24 hours.
Then follow these simple instructions:

  • Place your samples into micro tubes. Pack them in a suitable transport box. Fill with cooling units/cold packs (do not use dry ice!).
  • Print the PDF and stick it on the transport box or just hand it to your FedEx officer upon pickup.
  • Call to request a pickup from your local FedEx office.

If you plan to fill your transportation box with cooling packs, select this shipping option. If you plan to use dry ice, you cannot use this Waybill!

Get a quote for shipment by World Courier

The World Courier service can be used for shipment of samples of all types. If you select this option, we will contact you to provide a quotation based on location and shipping conditions. You will receive a prepaid Air Waybill which will be invoiced together with your analysis results. This is usually a good option if FedEx does not service your location.
